Did you put a $ before AMBERHOME?
Try
echo $AMBERHOME
and see if it points to /home/ayesha/amber/amber12
If so then $AMBERHOME/bin/tleap should work
Also try
echo $PATH
and see if it points to /home/ayesha/amber/amber12/bin
If so then you can just type tleap
